Each card exists only once in the deck, so holding it removes combos from your opponent’s range. Blockers are most useful when choosing bluffs: holding a card that blocks the nuts makes it less likely your opponent can call. They also help when deciding whether to call a big bet.

Example: On K♠ 8♠ 4♠ 2♦ 9♣, holding the A♠ means nobody else can have the ace-high flush. If you hold one ace, your opponent has only 3 combos of A-A instead of 6.
